    Initial Assessment of Acoustic Source Visibility with a 24-Element Microphone Array in the Arnold Engineering Development Center 80- by 120-Foot Wind Tunnel at NASA Ames Research Center

    Measurements of background noise were recently obtained with a 24-element phased microphone array in the test section of the Arnold Engineering Development Center 80- by120-Foot Wind Tunnel at speeds of 50 to 100 knots (27.5 to 51.4 m/s). The array was mounted in an aerodynamic fairing positioned with array center 1.2m from the floor and 16 m from the tunnel centerline, The array plate was mounted flush with the fairing surface as well as recessed in. (1.27 cm) behind a porous Kevlar screen. Wind-off speaker measurements were also acquired every 15 on a 10 m semicircular arc to assess directional resolution of the array with various processing algorithms, and to estimate minimum detectable source strengths for future wind tunnel aeroacoustic studies. The dominant background noise of the facility is from the six drive fans downstream of the test section and first set of turning vanes. Directional array response and processing methods such as background-noise cross-spectral-matrix subtraction suggest that sources 10-15 dB weaker than the background can be detected

    Advanced Background Subtraction Applied to Aeroacoustic Wind Tunnel Testing

    An advanced form of background subtraction is presented and applied to aeroacoustic wind tunnel data. A variant of this method has seen use in other fields such as climatology and medical imaging. The technique, based on an eigenvalue decomposition of the background noise cross-spectral matrix, is robust against situations where isolated background auto-spectral levels are measured to be higher than levels of combined source and background signals. It also provides an alternate estimate of the cross-spectrum, which previously might have poor definition for low signal-to-noise ratio measurements. Simulated results indicate similar performance to conventional background subtraction when the subtracted spectra are weaker than the true contaminating background levels. Superior performance is observed when the subtracted spectra are stronger than the true contaminating background levels. Experimental results show limited success in recovering signal behavior for data where conventional background subtraction fails. They also demonstrate the new subtraction technique's ability to maintain a proper coherence relationship in the modified cross-spectral matrix. Beam-forming and de-convolution results indicate the method can successfully separate sources. Results also show a reduced need for the use of diagonal removal in phased array processing, at least for the limited data sets considered

    Acoustic Study of a Sweeping Jet Actuator for Active Flow Control Applications

    Active flow control for aircraft has been demonstrated to improve aerodynamic performance but generally introduces additional noise sources. This study presents measurements of the acoustic fields of sweeping jet actuators at full- and half-scale compared to the configuration recently demonstrated in ground- and flight-tests of a full-scale rudder. The effects of scale, flow rate, forward speed, and actuator synchronization state on tone and broadband noise directional power spectral density (PSD) levels are presented. The sweeping jet actuator PSD levels in the audio range increases as the second power of actuator flow rate. A fourth power variation of PSD level with flow rate is observed for a comparable non-sweeping actuator. These measurements will be used to develop noise source models for new aircraft configurations utilizing active flow control

    Phased Acoustic Array Measurements of a 5.75 Percent Hybrid Wing Body Aircraft

    Detailed acoustic measurements of the noise from the leading-edge Krueger flap of a 5.75 percent Hybrid Wing Body (HWB) aircraft model were recently acquired with a traversing phased microphone array in the AEDC NFAC (Arnold Engineering Development Complex, National Full Scale Aerodynamics Complex) 40- by 80-Foot Wind Tunnel at NASA Ames Research Center. The spatial resolution of the array was sufficient to distinguish between individual support brackets over the full-scale frequency range of 100 to 2875 Hertz. For conditions representative of landing and take-off configuration, the noise from the brackets dominated other sources near the leading edge. Inclusion of flight-like brackets for select conditions highlights the importance of including the correct number of leading-edge high-lift device brackets with sufficient scale and fidelity. These measurements will support the development of new predictive models

The best tools for this purpose initially block export of the secretory cargo from the endoplasmic reticulum (ER), and then release the block to generate a cargo wave. However, previously developed regulatable secretory cargoes are often tricky to use or specific for a single model organism. To overcome these hurdles for budding yeast, we recently optimized an artificial fluorescent secretory protein that exits the ER with the aid of the Erv29 cargo receptor, which is homologous to mammalian Surf4. The fluorescentsecretory protein forms aggregates in the ER lumen and can be rapidly disaggregated by addition of a ligand to generate a nearly synchronized cargo wave. Here we term this regulatable secretory proteinESCargo (Erv29/Surf4-dependent Secretory Cargo) and demonstrate its utility not only in yeast cells, but also in cultured mammalian cells, Drosophila cells, and the ciliate Tetrahymena thermophila. Kinetic studies indicate that rapid export from the ER requires recognition by Erv29/Surf4.     Intestinal Interleukin-17 Receptor Signaling Mediates Reciprocal Control of the Gut Microbiota and Autoimmune Inflammation

    Interleukin-17 (IL-17) and IL-17 receptor (IL-17R) signaling are essential for regulating mucosal host defense against many invading pathogens. Commensal bacteria, especially segmented filamentous bacteria (SFB), are a crucial factor that drives T helper 17 (Th17) cell development in the gastrointestinal tract. In this study, we demonstrate that Th17 cells controlled SFB burden. Disruption of IL-17R signaling in the enteric epithelium resulted in SFB dysbiosis due to reduced expression of α-defensins, Pigr and Nox1. When subjected to experimental autoimmune encephalomyelitis, IL-17R signaling deficient mice demonstrated earlier disease onset and worsened severity that was associated with increased intestinal Csf2 expression and elevated systemic GM-CSF cytokine concentrations. Conditional deletion of IL-17R in the enteric epithelium demonstrated that there was a reciprocal relationship between the gut microbiota and enteric IL-17R signaling that controlled dysbiosis, constrained Th17 development, and regulated the susceptibility to autoimmune inflammation
